Abstract
Objective: To explore the optimal technology of celastrol extraction and purification from C.anticulatus Thunb.
Methods: By comparing the extraction efficiency of refluxing extraction, supercritieal C02 extraction and ultrasound-assisted extraction, ultrasound-assisted extraction was chosen eventually and optimized by orthogonal experiments; silica-gel column chromatography process for purification was also investigated.
Results: The optimal extraction process consisted of a liquid-solid ratio of l0 :1 ( v/w ), 400 W of extracting power, 5 seconds of once radiated time and 40 minutes of extraction time.The content of celastrol was not less than 90%, when purified by silica-gel column chromatography process.
Conclusions: The application of ultrasonic method showed to be highly efficient in the extraction of celastrol, compared with classical methods ; silica-gel column chromatography process was simple and the purification degree was high.
