Tröger's base analogs with diamide and sulfurous diamide bridges

We wish to report the synthesis of several novel analogs of Tröger's base (TB) 1. with amide and sulfurous diamide bridging units. Amide bridges were introduced onto the TB scaffold in one of three ways (Scheme 1): a) A diacid chloride was reacted directly with 1. to afford 3, b) The methylene bridge was removed from 1. and the resulting compounds of type 2 were reacted with a 1,2-diacid chloride or c) 2 was coupled with a 1,2-dicarboxylic acid via DCC coupling. Compounds of type 4 were afforded from the reaction of 2 with thlonyl chloride at low temperatures.
